Preferably in the position you will be using to make the cut.

It doesn't matter where the quill is, but it should be clamped.

I've read that the best position is with the quill down. I'm making my own tram tool from 5/16" dia. mild steel rod bent in an L position.
I'm making two, a shorter and a long one. The longer one will have a gusset welded in at the bend. This, plus a Mitutoyo back plunger.
The Starrett 196 is ok,....but a very old design. .001" for me., for mill tramming. I got this from Lautards MBSR.
